Présentation

Je pilote des projets de recherche portant sur les interactions immunobiologiques au sein des systèmes hôte/pathogène. Plusieurs objectifs guident ces projets : a) l’identification et de caractérisation des molécules clés de la virulence du pathogène et de la résistance de l’hôte, b) décortiquer les mécanismes de l’immunité chez les invertébrés et les replacer dans un contexte évolutif, c) lutter contre les infections relatives aux pathogènes étudiés. Nos approches expérimentales consistent principalement en des analyses comparatives d’expression de gènes et de protéines impliquées dans ces interactions en fonction de conditions environnementales changeantes, et cela tant au niveau des individus que des populations. Je suis également intéressé par la caractérisation fonctionnelle des protéines candidates ainsi identifiées. Je maitrise un large panel de techniques de biologie moléculaire et de biochimie. Pour la partie transcriptomique: extraction d’ADN, d’ARN et de protéines, PCR, clonage, mutagénèse dirigée, PCR quantitative, construction de banques EST, de banques de micorsatellites, de banques soustractives, hybridation *in situ*, mais également analyse de séquençage Sanger et de séquençage massif de transcriptome (RNA-seq), analyse de profil microsatellite en multiplex. Pour la partie protéomique : électrophorèse 1D de protéines, Western blot, immunoprécipitation, expression recombinante de protéines et leur purification, approches d’interactome. I am leading research projects dealing with immunobiological interactions in host/pathogen systems. Goals are a) the identification and characterization of key molecules involved in pathogen virulence and host resistance, b) to decipher the mechanisms of invertebrate immunity at the molecular and evolution levels, c) to fight against the disease caused by studied pathogens. Our experimental approaches are mainly comparative analyses of genes and proteins expression between different environmental or experimental conditions, at both individual and population levels. I am also interested by the functional validation of candidate protein identified. My expertise domain concern transcriptomic and proteomic approaches. At the transcriptomic level: nucleic acid extraction, PCR, cloning, mutagenesis, real time PCR, library construction (EST, microsatellite, substractive,…), in situ hybridization, RNA-Seq and Sanger sequencing analyses, microsatellite multiplex analyses. At the proteomic level: protein 1D electrophoresis, western blot, immunoprecipitation, heterologous and cell free expression system, protein purification system, interactome approach.
